Overview

Beta-actin-like protein 2 (ACTBL2), also known as kappa-actin, is a recently identified member of the actin protein family, often referred to as the seventh actin isoform. While it shares approximately 92% sequence similarity with beta-actin, it exhibits distinct biochemical properties and cellular localizations, primarily functioning in the regulation of the actin cytoskeleton and cell motility. ACTBL2 is notably upregulated in various malignancies, including colorectal cancer, ovarian cancer, melanoma, and hepatocellular carcinoma, where it promotes tumor cell proliferation, migration, and invasion. Its expression often correlates with poor patient prognosis and chemoresistance, making it a valuable diagnostic and prognostic biomarker. Mechanistically, ACTBL2 interacts with cytoskeletal regulators like gelsolin and competes for binding with metastasis suppressors such as MTSS1 to modulate oncogenic signaling pathways like NF-κB. Although no drugs targeting ACTBL2 are currently in clinical use, it is considered a promising therapeutic target, with experimental studies utilizing siRNA knockdown and small molecules like 5-arylidenethiazolidinone to inhibit its activity and reduce cancer cell viability.
