Overview

Beta-barrel assembly machinery protein A (BamA) is the essential, core component of the BAM complex responsible for the folding and insertion of beta-barrel outer membrane proteins (OMPs) in Gram-negative bacteria. Located in the outer membrane, BamA consists of a C-terminal transmembrane beta-barrel and multiple N-terminal polypeptide transport-associated (POTRA) domains that reside in the periplasm. It functions as a molecular machine that facilitates the thermodynamically challenging process of integrating proteins into the lipid bilayer, a process vital for maintaining the bacterial protective barrier and overall cell viability. Because BamA is highly conserved across Gram-negative species and lacks a human homolog, it has emerged as a high-priority target for novel antibiotics. Small molecules like darobactin and specialized monoclonal antibodies bind to BamA to lock its conformational state or block its lateral gate, effectively halting OMP biogenesis and causing rapid bacterial death. This mechanism is particularly effective against multi-drug resistant pathogens such as Acinetobacter baumannii and Pseudomonas aeruginosa.
