Overview

Catenin beta-1 (CTNNB1) is a dual-function protein involved in cell adhesion and Wnt signaling. It's a key component of adherens junctions, connecting cadherins to the actin cytoskeleton. In the Wnt pathway, it acts as a transcriptional coactivator for TCF/LEF transcription factors upon Wnt stimulation, promoting cell proliferation and differentiation. Dysregulation of CTNNB1, often through mutations preventing its degradation, is implicated in various cancers, making it an important oncogene. It also plays a role in cardiac disease. Targeting CTNNB1 therapeutically is challenging due to its critical roles in normal cellular processes.
