Overview

Beta-catenin-interacting protein 1 (CTNNBIP1) is a regulatory protein encoded by the human *CTNNBIP1* gene. Its main function is to bind β-catenin and prevent its interaction with TCF family transcription factors, thereby negatively regulating the Wnt/β-catenin signaling pathway. CTNNBIP1 is essential in controlling processes such as cell proliferation, migration, and gene transcription. It acts as a tumor suppressor in various cancers, particularly lung cancer and hepatocellular carcinoma, by inhibiting the transcriptional activation of β-catenin target genes like c-Myc, cyclin D1, and MMP7. Reduced CTNNBIP1 expression is associated with increased cancer cell migration, poorer prognosis, and higher expression of β-catenin target genes. CTNNBIP1 may serve as a prognostic biomarker in cancer, but there are currently no known drugs that directly target CTNNBIP1. It is not classified as a conventional receptor, enzyme, or transporter, but as a regulatory adaptor protein within the Wnt signaling cascade.
