The beta-catenin signaling pathway, often referred to as the canonical Wnt/β-catenin pathway, is a highly conserved cell signaling cascade crucial for regulating stem cell pluripotency, cell fate decisions during development, tissue homeostasis, and organogenesis. It also plays significant roles in tissue repair and is implicated in various diseases, including cancer. The pathway involves Wnt ligands, Frizzled receptors, LRP5/6, Dishevelled, a destruction complex (Axin, APC, GSK3β, CK1α), β-catenin, and TCF/LEF transcription factors. Aberrant activation or mutations affecting components can lead to excessive stabilization/nuclear localization of β-catenin, frequently observed in cancers such as colorectal carcinoma due to uncontrolled expression of proliferation-associated genes.
