Overview

Beta-crystallin A4 is a highly stable, evolutionarily conserved structural protein that forms part of the major protein complex in the vertebrate eye lens, encoded by the CRYBA4 gene. Belonging to the beta crystalline family (acidic subgroup), it contributes to lens transparency and refractive properties essential for vision. Mutations in CRYBA4 disrupt lens structure during development, leading to congenital cataracts, most commonly through autosomal dominant inheritance. The protein interacts with other beta-crystallins, forming heterodimers and larger aggregates to maintain lens integrity. There is currently no evidence that it serves as a conventional therapeutic target (e.g., receptor/enzyme/transport) for approved drugs, but its dysfunction is causally linked to hereditary ocular disease. Key notes: - Not a receptor, enzyme, or classical therapeutic target; its role is structural within the lens. - No known modulatory drugs in clinical use; no validated pharmacological targeting or safety profile as a drug target. - Genetic variants are involved in the pathogenesis of congenital and hereditary cataracts but are not used as biomarkers or direct therapeutic targets.
