Overview

Beta-DNA polymerase (pol β) is a small, essential eukaryotic enzyme primarily involved in the base excision repair (BER) pathway, which repairs damaged DNA bases and single-strand breaks. Pol β consists of an N-terminal dRP lyase domain and a C-terminal polymerase domain. It functions in gap-filling synthesis and removal of sugar-phosphate residues during BER. Inhibition of pol β can sensitize cells to chemotherapeutic agents that induce DNA damage, making it a potential therapeutic target, particularly in cancer.
