Beta-Klotho (β-Klotho, KLB) is an essential type-I single-pass transmembrane protein acting as a coreceptor for endocrine fibroblast growth factors, primarily FGF19 and FGF21. It is crucial for FGF19 and FGF21 signaling in metabolic tissues like the liver, adipose tissue, and pancreas, influencing energy homeostasis, glucose/lipid metabolism, and insulin sensitivity. It has also been implicated in tumor suppression.
Forms ternary complexes with FGFRs upon binding FGF19 or FGF21, enabling high-affinity binding and downstream signaling.
