Beta-lactamase class A enzymes are a major group of serine beta-lactamases produced by bacteria. They catalyze the hydrolysis of the beta-lactam ring found in beta-lactam antibiotics (such as penicillins and cephalosporins), rendering these drugs ineffective and conferring antibiotic resistance to the producing organisms. These enzymes are among the most clinically significant mechanisms of resistance, especially in Gram-negative pathogens. They utilize an active-site serine residue to attack the carbonyl carbon in the amide bond of beta-lactams, forming an acyl-enzyme intermediate and releasing inactive antibiotic. Genes encoding these enzymes are often located on mobile genetic elements, facilitating horizontal gene transfer and contributing to widespread antibiotic resistance.
