Beta-lactamase class A enzymes are a group of serine beta-lactamases produced by bacteria. These enzymes confer resistance to beta-lactam antibiotics (penicillins, cephalosporins) by hydrolyzing the beta-lactam ring. The active site includes key residues such as Ser70, Glu166 and Lys234. Inhibition of these enzymes is a strategy to combat antibiotic resistance, but mutations can lead to reduced inhibitor binding or expanded substrate range. Class A enzymes are a major cause of antibiotic resistance in Gram-negative bacteria. Inhibitors like clavulanic acid and avibactam are used to combat their activity.
