Beta-NGF is a neurotrophic factor and neuropeptide essential for the development, maintenance, proliferation, and survival of certain neuron populations. It exerts its effects by binding to two main classes of cell surface receptors: TrkA (high-affinity) and p75NTR (low-affinity), influencing neuronal survival, differentiation, and apoptosis depending on the context. Altered expression or signaling has been implicated in neurodegenerative diseases and neuropathic pain syndromes.
Binding to TrkA/NTRK1 stimulates neuronal survival, differentiation, axonal growth via activation of signaling pathways such as Ras/MAPK and PI3K/Akt. Binding to p75NTR can mediate both cell survival (via NF-kB pathway) or apoptosis (via JNK/c-Jun pathway), depending on co-receptor context and ligand form. proNGF preferentially binds to a complex of p75NTR with sortilin or SORCS2; this can induce apoptosis rather than promote survival.
