Beta-site APP-cleaving enzyme 1 (BACE1) is an aspartic protease that initiates the cleavage of amyloid precursor protein (APP) at the β-secretase site, leading to the production of amyloid-beta peptides (Aβ). This step is rate-limiting for Aβ production and is a key event implicated in Alzheimer's disease pathogenesis, making BACE1 a major drug target for AD therapy development. BACE1 also plays a role in myelin sheath formation.
