BH3-like motif-containing cell death inducer (BLID, also known as BRCC2) is a protein-coding gene encoding a 108-amino-acid protein containing a BH3-like motif that promotes apoptotic cell death via the mitochondrial/caspase pathway. BLID is broadly expressed in normal tissues but downregulated in breast and other cancers, correlating with poor prognosis and drug resistance. BLID acts downstream of the pro-apoptotic transcription factor FOXO3a and can sensitize cancer cells to chemotherapeutic agents such as doxorubicin and paclitaxel. Loss of BLID expression is associated with reduced apoptosis, enhanced proliferation, and increased metastasis in breast cancer models; restoration of BLID function suppresses growth and metastasis in vitro and in vivo. BLID is considered a tumor suppressor and a candidate biomarker for patient stratification in breast cancer and potentially other cancers[1][2][3].
