BHLHE40 Antisense RNA 1 (BHLHE40-AS1)

Target
BHLHE40-AS1
Molecular classification
Long non-coding RNA (lncRNA), Non-protein-coding RNA

Overview

BHLHE40 Antisense RNA 1 (BHLHE40-AS1) is a cytoplasmic, evolutionarily conserved human long non-coding RNA transcribed antisense to the BHLHE40 gene. It plays a significant role in cancer progression by promoting an immune-permissive microenvironment and supporting inflammatory signaling, notably through IL-6 and STAT3 pathway regulation. BHLHE40-AS1 is enriched in HER2-positive breast cancers and its expression correlates with increased invasiveness, decreased patient survival, and altered sensitivity to chemotherapy. Experimental reduction of BHLHE40-AS1 suppresses tumor cell growth and decreases inflammatory cytokine expression, indicating a functional role in tumorigenesis but not as a direct therapeutic target or receptor.

Mechanism of action

Not targeted by drugs directly. Mechanistically, may interact with IL-6 mRNA via ILF3 (an RNA-binding protein), support STAT3 pathway activation, and modulate inflammatory cytokine expression.

03

Biological functions

Regulation of inflammatory cytokine signaling (e.g., IL-6/STAT3)Modulation of immune-permissive microenvironment in cancerRegulation of protein expression, including DEC1/STAT3

Disease associations

05

Safety considerations

Not a drug target, thus direct safety concerns do not apply; however, aberrant expression is associated with poor prognosis and tumor aggressiveness.
06

Interacting drugs

Lapatinib—indirectly modulates expression in HER2+ breast cancer cells
