Overview

Bicaudal D cargo adaptor 1 (BICD1) is an evolutionarily conserved cytoskeletal motor adaptor protein that facilitates the loading and transport of intracellular cargo via the dynein/dynactin complex, playing crucial roles in microtubule organization, vesicular trafficking, and cell polarity. BICD1 is characterized by conserved coiled-coil domains facilitating cargo binding and interaction with molecular motors. It regulates the intracellular sorting and recycling of neurotrophin receptors (TrkB and p75^NTR^) in neurons, and modulates signaling by affecting receptor location and turnover, impacting neurotrophic factor responses and neuronal development. BICD1 also interacts with cellular signaling proteins such as PAR1, affecting the signaling pathways and promoting receptor endocytosis. Under hypoxic conditions, it is essential for HIF1α nuclear translocation. Its dysfunction has been implicated in diseases such as emphysema, otitis media, central corneal ulcer, and cancer progression. There is no evidence of BICD1 being a direct drug target or associated with approved therapeutics.
