Overview

The Bile canaliculus, often referred to as the canalicular anatomical lumen, is a microscopic, tube-like space formed by the apical membranes of adjacent hepatocytes (StatPearls, 2023). It represents the initial portion of the biliary system, serving as the primary site for the secretion of bile acids, cholesterol, and bilirubin (Wikipedia, 2024). This structure is an anatomical compartment rather than a molecular target such as a receptor or enzyme; however, it is the functional destination for various ATP-binding cassette (ABC) transporters, including the Bile Salt Export Pump (BSEP) and Multidrug Resistance-associated Protein 2 (MRP2) (NCBI, 2022). Proper maintenance of the canalicular lumen is essential for hepatic detoxification and the emulsification of dietary fats. Pathological conditions that obstruct or damage this lumen, such as primary biliary cholangitis or drug-induced cholestasis, lead to the systemic accumulation of toxic bile salts and potential hepatotoxicity (PubMed, 2021). While the lumen itself is not a direct drug target, its physiological integrity is a critical parameter in evaluating drug safety and biliary excretion profiles.
