Bacterial biofilm formation is a complex, multi-step process by which bacteria adhere to surfaces and develop structured communities encased in a self-produced extracellular polymeric substance (EPS) matrix. This process is central to bacterial survival in hostile environments and contributes significantly to antimicrobial resistance, chronic infections, and device-related complications. Biofilms provide bacteria with enhanced protection against environmental stresses, antibiotics, and the host immune system. The formation proceeds through reversible attachment, irreversible attachment, EPS production, maturation, and dispersal. Key regulatory mechanisms include quorum sensing and c-di-GMP signaling. Biofilms are implicated in numerous chronic infections due to their resilience against antibiotics/host defenses.
