Biorientation of chromosomes in cell division protein 1 (BOD1) is a small, conserved protein crucial for the accurate segregation of chromosomes during mitosis[1][4][6]. BOD1 localizes to spindle poles and kinetochores, where it plays an essential role in the establishment of correct microtubule–kinetochore attachments, ensuring sister chromatid biorientation and cohesion during cell division[1][4]. Its molecular function includes the inhibition of the protein phosphatase 2A (PP2A)–B56 complex, a key regulator of kinetochore substrate dephosphorylation[3][6]. Loss of BOD1 leads to defects such as syntelic microtubule attachments, elongated spindles, improper chromosome alignment, cohesion loss, and premature chromatid separation, resulting in genomic instability and eventual cell death[1][3][4][6]. While BOD1 is not currently considered a direct therapeutic target or a receptor/enzyme/transporter, its depletion or dysfunction can contribute to disease states, particularly cancers associated with chromosomal instability.
