Overview

Biorientation of chromosomes in cell division protein 1-like 1 (BOD1L1) is a component of the cellular fork protection machinery and plays a critical role in the maintenance of genome stability during cell division. Localized in the nucleoplasm, BOD1L1 participates in both the DNA damage response and the protection of stalled or damaged DNA replication forks from uncontrolled resection by nucleases. It acts through stabilization of RAD51 nucleofilaments at damaged forks, protecting these structures from anti-recombinogenic helicases such as FBH1 and BLM[3][6][7]. BOD1L1 is specifically required for efficient mitotic progression, faithful chromosome segregation, and proper regulation of kinetochore protein phosphorylation via interaction with the phosphatase PP2A and the kinase Aurora A[1]. Unlike its paralog BOD1, BOD1L1 does not regulate spindle orientation but is functionally distinguished by its role in DNA repair and mitotic fidelity[1][3]. Disruption of BOD1L1 leads to defects in chromosome alignment, abnormal spindle structures, and increased mitotic errors, which may contribute to chromosomal instability, a hallmark of many cancers[1]. No drugs or direct pharmacological agents are currently known to target BOD1L1, and it is not yet considered a therapeutic target in clinical practice.
