"Drains Fire" (Xie Huo) is a Traditional Chinese Medicine (TCM) therapeutic category and action focused on purging intense internal "fire" or pathogenic heat from the body [1, 2]. Modern pharmacological research has identified the Bitter Taste Receptor (TAS2R) family as a significant molecular target mediating this effect [7, 8]. These G protein-coupled receptors are expressed not only on the tongue for taste perception but also in extra-oral tissues such as the heart, lungs, and immune cells [3, 7]. In the heart, activation of TAS2Rs by bitter-tasting "fire-draining" compounds, such as berberine or baicalin, has been shown to decrease the contractile strength of the heart, providing a molecular basis for the traditional "cooling" and fire-draining effect [7, 8]. Furthermore, TAS2R activation on immune cells can suppress the release of pro-inflammatory cytokines like TNF-α and IL-6, assisting in the resolution of systemic inflammation associated with "fire" syndromes [11, 13].
Activation of extra-oral bitter taste receptors (TAS2Rs) by bitter ligands, which leads to the modulation of G-protein signaling pathways that decrease cardiac contractility and suppress the production of pro-inflammatory cytokines such as TNF-alpha and IL-6 [7, 13].
