The blood coagulation cascade components consist of a series of plasma proteins, predominantly zymogens and cofactors, that work in a highly regulated enzymatic sequence to maintain hemostasis. These factors, primarily synthesized in the liver, are activated through either the extrinsic or intrinsic pathways, which converge on the common pathway to generate the enzyme thrombin. Thrombin subsequently converts soluble fibrinogen into insoluble fibrin, forming a structural mesh that stabilizes the primary platelet plug and prevents hemorrhage. Deficiencies or mutations in these components lead to significant bleeding disorders such as hemophilia, while pathological over-activation can result in life-threatening conditions like deep vein thrombosis, pulmonary embolism, and stroke. Because of their central role in clot formation, individual components—most notably Factor Xa and thrombin—are primary therapeutic targets for a wide range of anticoagulant and antithrombotic drugs. Pharmacological modulation of the cascade remains a cornerstone of cardiovascular medicine for the prevention and treatment of thromboembolic events.
