CD303, also known as BDCA-2, is a type II transmembrane glycoprotein and a member of the C-type lectin superfamily. It is exclusively expressed on human plasmacytoid dendritic cells (pDCs) and plays a crucial role in antigen capture and the negative regulation of type I interferon production. CD303 signals via association with the FcεRIγ adaptor protein, leading to the recruitment and phosphorylation of spleen tyrosine kinase (SYK). Targeting CD303 is being explored as a therapeutic strategy for autoimmune diseases and cancer.
Antibody targeting against CD303 can suppress excessive type I interferon production by crosslinking, engagement, or through complement-dependent cytotoxicity.
