Blood Restricted Antigen 3 (BRA3) is a lineage-restricted antigen presented as a peptide in the context of Human Leukocyte Antigen (HLA), specifically HLA-A*02:01. This peptide-MHC complex is predominantly expressed on cells of the hematopoietic lineage, including both malignant leukemia cells and healthy hematopoietic stem and progenitor cells. Because its expression is restricted to the blood-forming system, it serves as a highly specific target for cellular therapies like TCR-T cells in the treatment of hematologic malignancies such as acute myeloid leukemia (AML). Targeting BRA3 allows for the elimination of leukemic blasts, though it necessitates subsequent hematopoietic stem cell transplantation to restore the healthy blood system due to the depletion of normal hematopoietic cells. Therapeutic candidates like LYL132 are engineered to recognize this specific complex to induce potent anti-tumor immunity.
