Overview

BMERB domain-containing protein 1 (BMERB1) encodes a protein characterized by the bMERB domain. This domain is a bivalent Mical/EHBP Rab binding motif, functioning as a Rab8 family effector evolved by gene duplication. The BMERB domain enables the protein to bind with high specificity to Rab8 family small GTPases, including Rab8, Rab10, Rab13, and Rab15, which are key regulators of endosomal trafficking. BMERB1 is predicted to be involved in the negative regulation of microtubule depolymerization and cell motility, especially in radial glia-guided migration in the cerebral cortex. It is expressed in several tissues, including the central nervous system. The protein is not recognized as a therapeutic target, nor are there known drugs or established biomarker roles for it. Disease associations according to genetic databases include Noonan Syndrome 10 and Lynch Syndrome 1. The molecular architecture and evolutionary history of the bMERB domain indicate specialized cellular trafficking and cytoskeletal roles, but its detailed physiological and pathological implications remain under investigation.
