BMP-2-inducible protein kinase (BMP2K) is a serine/threonine protein kinase implicated in the regulation of osteoblast differentiation. It is part of the Numb-associated kinase (NAK) family, which have roles in diverse cellular processes including endocytosis and modulation of developmental signaling pathways. BMP2K expression is upregulated during BMP-2-induced differentiation, and the protein is thought to act as a negative regulator in bone formation by attenuating osteoblast differentiation. There are at least two transcript variants encoding different isoforms. Structural studies have confirmed its kinase fold and suggest potential druggability, but no selective therapeutic agents are currently approved that specifically target BMP2K. Its role in disease remains underexplored, but related NAK kinases are linked to cancer, neurodegeneration, and viral infection.
Putative: kinase inhibitors (inhibiting BMP2K or related NAK kinases can block phosphorylation-dependent signaling). No marketed drugs with an explicit mechanism targeting only BMP2K noted; investigational compounds targeting the NAK family exist.
