Bone extracellular matrix proteins are a diverse group of structural and regulatory proteins secreted primarily by osteoblasts, the bone-forming cells. The bone ECM consists of about 70% inorganic mineral (hydroxyapatite) and 30% organic matrix, the latter dominated by type I collagen (~90%) and smaller amounts of non-collagenous proteins (e.g., osteonectin, osteopontin, bone sialoprotein, proteoglycans, and glycoproteins)[2][3][7]. These proteins serve as the scaffold for mineral deposition, mediate cell-matrix interactions critical for osteoblast adhesion and differentiation (mainly via integrins), and regulate bone tissue’s mechanical properties and signaling for growth, repair, and homeostasis[2][3][1][5][6]. Osteoblasts deposit bone matrix, which then mineralizes to form mature bone; they also secrete factors that influence the differentiation and activity of other bone-resident cells, such as osteoclasts. The dysfunction of bone extracellular matrix proteins or osteoblasts is implicated in diverse skeletal pathologies such as osteoporosis, bone fractures, and inherited bone disorders[2][3][7][6].
