The bone marrow hematopoietic niche is a complex microenvironment composed of cellular (e.g., mesenchymal stromal cells, endothelial cells, osteoblasts, nerve fibers), molecular, and extracellular matrix components that maintain, regulate, and protect hematopoietic stem cells (HSCs) throughout life. This niche regulates stem cell fate decisions—including quiescence, self-renewal, proliferation, differentiation, homing, and mobilization—via numerous cell–cell interactions and signaling molecules. The integrity and proper functioning of the niche are essential for healthy blood formation; disruption by stress, chemotherapy, or aging leads to degenerative changes, impaired stem cell function, and increased disease risk, including hematological malignancy.
