Overview

Bone metastatic lesion" refers to an area of bone affected by metastasis, meaning cancer has spread from its original site to the bone. This is not a molecule, protein, or receptor but rather a radiological or pathological description of a region of bone invaded by cancer cells. Bone metastases most commonly originate from cancers of the breast, prostate, lung, kidney, and thyroid. Lesions are categorized as osteolytic (bone breakdown), osteoblastic (abnormal bone formation), or mixed, reflecting the disrupted bone remodeling caused by the invading tumor. Underlying molecular mechanisms involve factors such as RANK/RANKL, PTHrP, CXCR4, various integrins, and growth factors, but these are features of the biology of metastasizing cells rather than of the "lesion" itself. The presence of bone metastatic lesions is associated with pain, increased risk of fractures, spinal cord compression, and hypercalcemia, all contributing to significant morbidity[1][2][3][5][7]. Note: "Bone metastatic lesion" is not a therapeutic target, molecule, or receptor. It describes a pathological phenomenon (metastasis to bone) and thus is not a molecular entity suitable for drug targeting in the traditional sense, though many therapies aim to treat the effects or underlying biology of these lesions.
