Overview

Bone mineral at sites of increased osteogenesis in metastatic lesions refers to the mineralized matrix deposited during abnormal bone formation, typically seen in osteoblastic (bone-forming) metastases. These are most commonly associated with cancers such as prostate and breast cancer that have spread to bone. The process involves accelerated but often disorganized deposition of new bone matrix, resulting in areas with altered mineral content and structure compared to healthy bone. This newly formed bone is frequently poorly mineralized or has abnormal crystal structure and composition compared to normal bone tissue, detectable by imaging modalities sensitive to active ossification/mineralization. This is driven by tumor-secreted factors stimulating osteoblasts, disrupting normal homeostasis, and activating pathways like Wnt/β-catenin signaling. Despite increased density on imaging, these regions are often mechanically weaker and prone to fracture.
