Bone morphogenetic protein 8B (BMP8B)

Target
BMP8B
Molecular classification
Growth factor, Cytokine, Ligand of the TGF-β receptor family, Member of the TGF-β superfamily
01

Overview

Bone morphogenetic protein 8B is a secreted growth factor of the TGF-β superfamily, involved in embryonic development, bone and cartilage formation, cell differentiation, and tissue regeneration. It shares strong sequence homology with BMP7 and BMP5, and exerts its effects via binding to cell-surface TGF-β receptors, activating Smad-mediated intracellular signaling. BMP8B has been implicated in brown adipose tissue thermogenesis and may play roles in skeletal disorders, wound healing, and muscle regeneration. Recombinant BMP8B is under investigation for use in regenerative medicine, but no drugs directly target BMP8B clinically. Safety data for BMP8B are limited, but general concerns include potential ectopic bone formation and immunogenicity.

Mechanism of action

Drug candidates or recombinant BMP8B would act by binding to TGF-β receptors, activating Smad signaling, resulting in the induction of bone/cartilage formation, tissue regeneration, and modulation of cell fate and differentiation
