Overview

Bone morphogenetic protein receptors (BMPRs) are transmembrane serine/threonine kinase receptors that mediate the cellular effects of bone morphogenetic proteins (BMPs), members of the transforming growth factor-beta (TGF-β) superfamily. BMPs are secreted cytokines involved in a wide range of biological processes, including embryonic development, cell differentiation, proliferation, apoptosis, and especially bone and cartilage formation. BMPRs function as heteromeric complexes composed of type I (ALK2/ACVR1, ALK3/BMPRIA, ALK6/BMPRIB) and type II (BMPRII) receptors. Upon ligand binding, the receptors phosphorylate SMAD proteins to regulate gene expression relevant to osteogenesis and other developmental processes. Disruptions or mutations in BMPR genes can lead to developmental disorders or diseases such as pulmonary arterial hypertension or impaired bone growth.
