BOS complex subunit NOMO1 (NOMO1) is a type I transmembrane glycoprotein located in the endoplasmic reticulum (ER) that plays a critical structural role in maintaining ER sheet morphology[1][2][3]. NOMO1 is a widely conserved protein found in both animals and plants, forming part of the multi-pass translocon (MPT) complex, which is involved in the insertion of multi-pass membrane proteins into cell membranes[2][5][6]. Structurally, NOMO1 is characterized by a rod-shaped architecture with multiple immunoglobulin-like (Ig-like) domains arranged in a “beads on a string” configuration, consistent with a flexible scaffold that helps define the ER's intermembrane spacing[1]. Overexpression of NOMO1 constrains the ER sheet luminal diameter, whereas depletion causes collapse and disruption of ER morphology. Although originally associated with nodal signaling pathways during development, its main documented function is structural rather than as a traditional druggable target such as receptor, enzyme, or transporter[1][3][5]. NOMO1 is not currently considered a direct therapeutic target, and no drugs are known to specifically interact with or modulate this protein[5][6].
