Overview

Bothrops spp. serine proteases (SVSPs) are a major family of enzymes found in the venom of pit vipers within the Bothrops genus, such as Bothrops jararaca and Bothrops asper. These enzymes play a critical role in the pathophysiology of envenomation by disrupting the host's hemostatic system, often acting as thrombin-like enzymes that cleave fibrinogen into unstable fibrin clots, leading to consumptive coagulopathy (UniProt P04971; PMID: 30145210). Beyond coagulation, they can influence the complement system and blood pressure regulation by releasing kinins or activating various plasma proteins (PMID: 25843415). In a clinical context, these proteases are the primary targets for therapeutic antibodies found in antivenoms, which aim to neutralize their enzymatic activity to prevent systemic hemorrhage and tissue necrosis. While some specific SVSPs like batroxobin have been repurposed as pharmacological tools for defibrinogenation, the broader class remains a significant challenge in toxicology due to its high functional diversity and isoform variation across different snake species (PMID: 33561455).
