Botulinum neurotoxin serotype E (BoNT/E) is one of the seven major serotypes of botulinum neurotoxins produced by Clostridium species. BoNT/E is a primary cause of human botulism, a potentially lethal paralytic disease that targets cholinergic nerve terminals. It is synthesized as an inactive single-chain polypeptide, which becomes active after proteolytic cleavage into a heavy and light chain. The light chain is a zinc-dependent metalloprotease that cleaves SNAP-25, a SNARE protein essential for acetylcholine release, leading to flaccid paralysis. While highly toxic, controlled doses are used therapeutically to treat certain neuromuscular disorders due to its ability to induce temporary muscle relaxation.
