Overview

BPI fold-containing family B member 1 (BPIFB1) is a secreted protein belonging to the BPI/LBP/PLUNC superfamily, characterized by the BPI fold structural motif which is involved in binding hydrophobic and amphipathic molecules, crucial for mucosal immunity[1][4]. BPIFB1 is expressed mainly in the mucosa of the mouth, nasal cavities, lungs, and other epithelial surfaces[1][4]. Its biological role centers on innate immune defense against Gram-negative bacteria and modulation of inflammation[1]. Clinically, BPIFB1 is dysregulated in various diseases including respiratory inflammatory diseases and several cancers, such as nasopharyngeal carcinoma (where it acts as a tumor suppressor by inhibiting cell migration, invasion, and metastasis through interference with the VTN/ITGAV/FAK/Src/ERK signaling pathway[2]) and hormone receptor-positive breast carcinoma (where it appears to promote metastasis via modulation of macrophage M2 polarization[3]). Expression of BPIFB1 is therefore both a potential biomarker for disease prognosis and a candidate therapeutic target, although no drugs are currently approved to directly modulate its activity.
