Overview

B-Raf V600E refers to a specific mutation in the BRAF gene, where the amino acid valine (V) at position 600 is replaced by glutamic acid (E). This single-point mutation results in a constitutively active form of the B-Raf protein, a serine/threonine kinase involved in cell signaling pathways that regulate cell growth and division. The mutant protein can signal independently of upstream Ras activation, leading to continuous MAPK pathway activity, driving uncontrolled cellular proliferation and survival. It is a therapeutic target in multiple cancer types and can be detected through DNA sequencing and immunohistochemistry.
