Overview

The brain-derived neurotrophic factor receptor refers to the cell surface receptors that bind and mediate the effects of brain-derived neurotrophic factor (BDNF). The two main receptors are TrkB, a high-affinity tyrosine kinase receptor, and p75NTR/LNGFR, a low-affinity pan-neurotrophin receptor. TrkB activation promotes neuronal survival, differentiation, and synaptic plasticity, while p75NTR can induce apoptosis or neurite outgrowth depending on the cellular context. Dysregulation of these receptors is implicated in neurological diseases.

Mechanism of action

Mature BDNF binds to TrkB, activating intracellular signaling cascades (PI3K/Akt, MAPK/ERK, PLCγ) promoting neuronal survival and plasticity. ProBDNF and mature BDNF can bind to p75NTR, triggering apoptosis or neurite outgrowth depending on cellular context and co-receptors.
