Overview

Brain enriched myelin associated protein 1 (BCAS1) is a multifunctional protein found in both the brain and various tumors. In the central nervous system, BCAS1 is a marker of early myelinating oligodendrocytes and immature oligodendrocyte lineage cells, with high expression during active myelination. In oncology, BCAS1 is overexpressed or amplified in several aggressive cancer types, including breast, prostate, pancreatic, colorectal, and gastric cancers, and its presence correlates with cancer cell proliferation, migration, and poor prognosis. BCAS1 also affects resistance to targeted therapies (e.g., cetuximab in colorectal cancer, tamoxifen in breast cancer) and can be modulated by epigenetic drugs. Its dual roles suggest importance both as a diagnostic/prognostic biomarker and as a potential therapeutic target, though targeting poses safety risks due to its functions in the nervous system and cancer biology[1][2][3][4].

Mechanism of action
