Overview

Brain-expressed X-linked protein 2 (BEX2) is a member of the BEX gene family, encoding a small adaptor protein implicated in the development of the nervous system and in cancer biology. BEX2 interacts with transcription factors such as LIM domain only 2 (LMO2) and is involved in transcriptional regulation by DNA-binding complexes. It plays highly context-dependent roles in cancer, acting as a pro-oncogenic factor that drives cell proliferation, survival, migration, and invasion, particularly through modulation of the NF-κB and JNK/c-Jun pathways in breast, hepatocellular, colorectal cancers, and various brain tumors. In neural tissue, BEX2 is essential for normal development, and knockout studies in mice show wide-ranging transcriptional and physiological effects. BEX2 is being investigated as an oncogenic driver and a potential therapeutic target in malignancies characterized by BEX2 overexpression. No approved therapeutics yet directly target BEX2, and the full safety profile for BEX2 inhibition remains uncharacterized.
