Overview

BRCA1-A complex subunit Abraxas 1 (ABRAXAS1) is a central scaffold protein within the BRCA1-A complex, functioning as an adaptor that links BRCA1 to other complex members such as RAP80, MERIT40, BRCC45, and BRCC36[2][3]. ABRAXAS1 is crucial for orchestrating the recruitment of BRCA1 to DNA double-strand breaks, thereby enabling effective DNA damage checkpoint activation and homologous recombination repair[1]. Germline and somatic mutations, as well as loss of function or reduced expression of ABRAXAS1, are associated with the development of several human cancers, mainly due to impaired DNA repair and loss of genome stability[1][2]. Despite its role in pathways intimately linked to cancer and DNA repair, ABRAXAS1 is not itself currently viewed as a direct therapeutic target or receptor, enzyme, or transporter. Rather, it is a tumor suppressor and DNA repair adaptor protein whose loss or mutation contributes indirectly to cancer pathogenesis[1][2][3].
