BRCA1-A complex subunit MERIT40 (BABAM1) is a core component of both the BRCA1-A deubiquitinase complex and the BRISC complex, which play pivotal roles in the cellular response to DNA double-strand breaks. MERIT40 stabilizes the BRCA1-A complex by mediating interactions with other core proteins such as RAP80, Abraxas, BRCC36, and BRE, facilitating the recognition and processing of Lys63-linked polyubiquitin chains at damaged chromatin sites. This is essential for the proper recruitment and retention of BRCA1 to DNA lesions, thereby ensuring chromatin remodeling, checkpoint activation, and choice of DNA repair pathway. Dysfunction or mutation of BABAM1 is associated with defective DNA damage repair and has been linked to increased susceptibility to breast and ovarian cancers due to impaired genomic integrity[1][2].
