Overview

BCR-ABL is a fusion protein with constitutive tyrosine kinase activity, resulting from the translocation between chromosomes 9 and 22, known as the Philadelphia chromosome. This fusion gene is a hallmark of chronic myelogenous leukemia (CML) and some other leukemias. BCR-ABL has constitutive tyrosine kinase activity, which leads to uncontrolled cell proliferation. It phosphorylates itself and numerous downstream signaling proteins, disrupting normal cellular processes such as apoptosis, differentiation, and adhesion—driving leukemogenesis.
