Overview

Bridge-like lipid transfer protein family member 2 (BLTP2) is an evolutionarily conserved lipid transfer protein that forms a bridge-like conduit for lipid transport between organelles, particularly between the endoplasmic reticulum (ER) and other membranes such as endosomes and the plasma membrane[1][2][4]. BLTP2 belongs to the VPS13-family, characterized by a long tubular structure with a hydrophobic core that facilitates bulk lipid movement. In human cells, BLTP2 is enriched at ER tubules and ER-endosome contact sites, where it regulates organelle lipid homeostasis and modulates plasma membrane fluidity by maintaining phosphatidylethanolamine (PE) levels[2]. Functionally, BLTP2 acts as a negative regulator of ciliogenesis and supports cell survival and proliferation, and its overexpression is correlated with poor prognosis and increased metastasis in breast cancer[1][2]. Loss-of-function studies in model organisms show that BLTP2 is critical for viability and secretion processes, highlighting its fundamental cellular roles[1][4]. There are currently no known approved drugs that directly interact with this protein, but its role in cancer biology makes it a potential future therapeutic target.
