Overview

Broad adaptive immune activation refers to a pharmacological mechanism where a therapeutic agent, typically a vaccine, induces a systemic immune response rather than interacting with a single, discrete host molecular target (IUPHAR/BPS, 2024). This process involves the coordinated action of the innate and adaptive immune systems, where antigens are captured by dendritic cells and presented to naive T and B lymphocytes (StatPearls, 2023). The resulting immune cascade leads to the production of antigen-specific antibodies and the generation of cellular immunity, providing long-term protection against pathogens or tumor cells (CDC, 2023). Unlike targeted therapies that modulate specific proteins, this approach leverages the entire immune network to achieve a therapeutic effect. Because the efficacy of these agents is mediated through complex cellular interactions and signaling pathways across multiple tissues, they are classified as having no discrete molecular target. This mechanism is the foundation of immunization and is increasingly utilized in the development of therapeutic cancer vaccines.
