Bromo adjacent homology domain-containing protein 1 (BAHD1) is a chromatin-associated protein that acts as a scaffold for a corepressor complex involved in epigenetic gene silencing [1, 2]. By recruiting histone modifiers such as deacetylases and methyltransferases, BAHD1 facilitates the formation of heterochromatin, thereby regulating the transcription of genes involved in the immune response and metabolism [2, 4]. It is notably recognized for its role in the type III interferon response, where it serves as a target for bacterial effectors that seek to modulate host immunity [2]. In oncology, BAHD1 is often upregulated and associated with the silencing of tumor-suppressive pathways, particularly in colorectal cancer [3]. Targeting BAHD1 mRNA via antisense oligonucleotides or RNA interference offers a method to reduce BAHD1 protein levels, potentially restoring normal gene expression in cancer or enhancing immune defense against pathogens [1, 3]. However, the broad regulatory role of BAHD1 across the genome presents challenges regarding the specificity and safety of such therapeutic interventions [2, 4].
