Bromodomain and extra-terminal domain (BET) proteins are a family of epigenetic "reader" proteins that play a central role in the regulation of gene transcription. They are characterized by the presence of two tandem N-terminal bromodomains (BD1 and BD2) and a conserved extra-terminal (ET) domain. BET proteins bind to acetylated lysines on histone tails, thereby regulating chromatin structure and gene expression during cellular proliferation and differentiation. BET family dysfunction is implicated in several diseases, including cancer and viral infection.
