Overview

Bromodomain and WD repeat-containing protein 3 (BRWD3) is a multi-domain scaffold protein harboring bromodomains and multiple WD repeats, which confer the ability to interact with acetylated and methylated histones, respectively[1][3][4][7]. BRWD3 acts as a chromatin-modifying factor and functions as a substrate-specificity receptor for the Cul4-DDB1 E3 ubiquitin ligase complex, thereby regulating the ubiquitin-mediated degradation of proteins on chromatin[1]. It directly regulates the stability of the histone demethylase KDM5, promoting its ubiquitination and degradation to maintain balanced levels of H3K4 methylation, a key histone modification linked with active gene transcription[1]. BRWD3 is essential for proper chromatin structure, gene expression, and cellular morphology, and its dysfunction is associated with X-linked intellectual disability, macrocephaly, and chromosomal abnormalities in certain leukemias[4][5][7]. There are currently no known drugs that directly target BRWD3 in the clinic or in preclinical studies.
