Bromodomain testis-specific protein (BRDT) is a member of the Bromodomain and Extra-Terminal (BET) family of epigenetic readers, primarily expressed in the testes during spermatogenesis (UniProt Q9NPI1). The first bromodomain (BD1) of BRDT specifically recognizes and binds to acetylated lysine residues on histone tails, particularly histone H4, which is essential for proper chromatin remodeling and gene expression during sperm development (Gaucher et al., 2012). Because of its restricted expression pattern, BRDT-BD1 is a highly attractive target for the development of non-hormonal male contraceptives (Matzuk et al., 2012). Small molecule inhibitors that block the BD1 pocket prevent BRDT from binding to chromatin, leading to a reversible reduction in sperm production and motility. Additionally, BRDT is classified as a cancer-testis antigen, as it is aberrantly expressed in various malignancies, including lung and squamous cell carcinomas, making it a potential target for epigenetic-based cancer therapies (Scanlan et al., 2004). Therapeutic challenges include achieving selectivity over other BET family members like BRD4 to avoid systemic side effects such as thrombocytopenia. Research into BD1-selective inhibitors aims to minimize these off-target effects while maintaining contraceptive efficacy.
