Transcription regulator protein BACH2 is a member of the basic leucine zipper (bZip) transcription factor family, characterized by a BTB/POZ domain and a bZip_Maf domain. It plays crucial roles in adaptive immunity by repressing terminal differentiation of lymphocytes, promoting long-lived memory T cells, supporting regulatory T cell and B cell development, and preventing excessive inflammatory or autoimmune responses. BACH2 controls gene expression by binding to Maf recognition elements and interacting with AP-1 family transcription factors, acting as a checkpoint for immune cell fate. Loss or mutation of BACH2 has been linked to immunodeficiencies (notably IMD60), autoimmunity, and cancers of the lymphoid lineage[1][3][4][5].
Transcriptional repression or activation via binding to Maf recognition elements (MAREs); impacts immune cell differentiation and function by regulating gene expression in T and B lymphocytes[1][5].
