Overview

BTB domain-containing protein 18 (BTBD18) is a protein primarily expressed in the testis, and is specifically required during spermatogenesis to promote the transcription of precursor piRNAs[1][3][5][7]. It acts in the nucleus, facilitating transcription elongation at piRNA loci during male meiosis, which is critical for the repression of transposable elements in germ cells[1][5][7]. Mouse deficiency studies indicate a key requirement for BTBD18 in spermiogenesis, as knock-out leads to increased germ cell apoptosis and male infertility[9]. There is no evidence that BTBD18 serves as a pharmacological target (e.g., receptor, enzyme) or is currently drugged; it is a structural and regulatory protein in the BTB/POZ protein family, which is known for protein-protein interactions and gene regulatory roles[2][4][6].
